An increase in uteroplacental blood flow is required to supply oxygen and nutrients to the growing conceptus. However, mechanisms which enhance uterine vascular growth during gestation are
poorly understood. In the present study, pairs of uterine arteries from late-pregnant rabbits (n=6) were dissected - one fed a placenta (PL) and one fed the uterine wall (UT); structural and
functional differences compared. Arteries were mounted on two glass micro-cannula in a specialized arteriograph chamber that allowed for control over transmural pressure (TMP) and
measurement of lumen diameter. When TMP was increased step-wise from 25→125mmHg both artery types actively constricted and developed pressure-dependent tone, however, UT arteries had
significantly more tone than PL arteries over the entire pressure range:%tone@50, 75, 100, 125mmHg for PL vs. UT=20±4 vs. 29±5, 31±2 vs. 39±2, 36±2, vs. 42±2, 36±2, vs. 45±3;(p<0.01).
Under fully relaxed conditions (papaverine treated), the PL arteries were 26% larger than UT arteries: φPL=168±17μm vs. φUT=124±10μm (p<0.05), but were significantly less distensible @
TMP=25-125mmHg (p<0.05), indicating differences in both cellular growth and extra-cellular matrix remodeling of the vascular wall for the two vessel types. These structural differences
are likely to contribute to differential regulation of placental versus myoendometrial flow. These data also suggest that a localized factor(s) contributes to the greater gestational
